Evaluation of antibacterial properties of alcohol and water extracts of propolis on Ralstonia solanacearum

Abstract
Potato brown rot (the causal agent Ralstonia solanacearum) is one of the important and dangerous bacterial diseases. Due to its wide distribution and broad host range and in soil of different regions through irrigation water or latent infected tubers. It is generally difficult to control the damage of of this bacterium. This disease is widely distributed in most part of Iran and which reduce the quality and quantity of potato. This research was conducted to survey antibacterial activity of propolis on Ralstonia solanacerum. After propolis collection from different parts of Kurdistan province and propolis extractionand water extraction, Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) activity for bacterial strains were determined. Data analysis was performed using the SPSS software programme and comparison of means was done with Duncan test at 5% level. Use of alcoholic solvent (96% ethanol and dimethyl sulfoxide) resulted in a greater mean diameter of growth inhibitory zone in comparison to water extract (p<0.05). Inhibitory concentrations (MICs) of alcoholic, dimethyl sulfoxide and water extract of propolis for Ralstonia solanacearum were 0/656, 1/31 and 2/62 and and the MBCs for each of the above mentioned extraction were 0/656, 1/31 and 5.25 mg/ml respectively. This disease by causing wilt and brown rot in potato region, cause serious disease in most potato growing region of Iran. This research for control of this disease conducted. According to the results, Propolis showed significant effect on Ralstonia solanacearum in laboratory condition.
ont-�s:2� �nt-family:"Times New Roman","serif";mso-bidi-font-family:"B Zar"'>SPSS و آزمون آماری DUNCAN استفاده گردید. میانگین قطر هاله عدم رشد عصاره الکلی (با استفاده از دو حلال اتانول 70 درصد و دی متیل سولفواکسید) به صورت معنیداری بیش از عصاره آبی بره موم بود ((p<0/05. حداقل غلظت مهارکنندگی (MIC) عصاره الکلی اتانولی، دی متیل سولفوکسید و عصاره آبی بره موم برای باکتری بیماری زای گیاهی Ralstonia solanacearum به ترتیب برای 656/0، 31/1 و 62/2 میلی گرم بر میلی لیتر و همچنین حداقل غلظت کشندگی (MBC) برای باکتری مذکور به ترتیب 656/0، 31/1 و 25/5 میلیگرم بر میلیلیتر تعیین گردید. از آن جا که این بیماری با ایجاد پژمردگی و پوسیدگی قهوه ای به عنوان خطری بالقوه ، محصول سیب زمینی کشور را تهدید می کند و مبارزه با آن مشکل است، بنابراین این پژوهش که برای کنترل این بیماری انجام پذیرفت و نشان داد که عصاره الکلی و آبی بره موم قادر به کنترل باکتری مذکور در شرایط آزمایشگاهی است.
